The translation initiation factor EIF2 catalyzes the first regulated step of protein synthesis initiation, promoting the binding of the initiator tRNA to 40S ribosomal subunits. Western blot analysis of extracts of various cell lines, using Phospho-eIF2α-S51 Rabbit MAb at 1:1000 dilution. HeLa and NIH/3T3 cells were treated by Calyculin A (100 nM) at 37℃ for 30 minutes after serum-starvation overnight or treated by TNF-α (20 ng/mL) at 37℃ for 30 minutes.
Secondary antibody: HRP Goat Anti-Rabbit IgG (H+L) at 1:10000 dilution.
Lysates/proteins: 25ug per lane.
Blocking buffer: 3% nonfat dry milk in TBST.
Detection: ECL Basic Kit.
